How do you use a GasPak?
How do you use a GasPak?
Place inoculated agar plates in an anaerobic jar, container or pouch. Add anaerobic indicator strip. Add the required number of gas generator sachet after opening the sealed GasPak container and place the lid in place or seal the pouch/container. Incubate at the desired temperature.
How do anaerobic sachets work?
It is used to culture bacteria which die or fail to grow in the presence of oxygen (anaerobes). These are commercially available, disposable sachets containing a dry powder or pellets, which, when mixed with water and kept in an appropriately sized airtight jar, produce an atmosphere free of elemental oxygen gas (O2).
How do you make an anaerobic jar?
Procedure
- Keep the inoculated culture plates inside the jar along with an indicator.
- Screw tight the lid.
- Close the inlet tube and connect outlet tube to a vacuum pump ( at least three quarters of the air of the jar can be removed).
What does the GasPak do?
GasPak System The GasPak Anaerobic System is used to create an oxygen-free environment for the growth of anaerobic microorganisms. Inoculated plates or tubes are placed inside the chamber, and anaerobic conditions are created by adding water to a gas generator envelope that is placed in the jar just before sealing.
What does anaerobic bacteria need to grow?
oxygen
An anaerobic organism or anaerobe is any organism that does not require molecular oxygen for growth. It may react negatively or even die if free oxygen is present. In contrast, an aerobic organism (aerobe) is an organism that requires an oxygenated environment.
What is anaerobic jar used for?
McIntosh and Filde’s anaerobic jar is an instrument used in the production of an anaerobic environment. This method of anaerobiosis as others is used to culture bacteria which die or fail to grow in presence of oxygen (anaerobes).
What component in the GasPak 100 removes oxygen from the jar?
The hydrogen gas combines with free oxygen in the chamber to produce water, removing all free oxygen from the chamber. This reaction is catalyzed by the element palladium, which is attached to the underside of the lid of the jar.
Can Microaerophiles grow in GasPak?
Many microaerophilic bacteria will grow poorly at 22% O2, whereas some will not grow at all (e.g. Neisseria gonorrhoea). The Gram+ genus Bacillus and Gram– genus Pseudomonas include aerobic bacillus-shaped bacteria. On the left is a GasPak jar, with a gas generator envelope inside. The environment is 0% O2,.
